Oxidation of Phenylpyruvate by Sweetclover Peroxidase
نویسندگان
چکیده
Sweetclover (Melilotus alba Desr.) contains a soluble enzyme which oxidizes phenylpyruvate with the addition of either a natural cofactor or Mn2+ and dichlorophenol. The final product of oxidation has not been identilied. Phenylpyruvate oxidation is catalyzed also by horseradish peroxidase, and the oxidation is enhanced by the natural cofactor from sweetclover or Mn2+ and dichlorophenol. After fractionation of sweetclover homogenates by DEAE-cellulose chromatography, two peaks of peroxidase.activity (pyrogallol as substrate) are apparent; both peaks also oxidize phenylpyruvate. A third peak, specific for phenylpyruvate oxidation, also is apparent. The Culcu and Blb allelic pairs, which influence phenolic metabolism in sweetclover have little, if any, influence on peroxidase activity. An autoxidation of phenylpyruvate occurs at high MnZ+ concentrations.

Oxidase-peroxidase enzymes of Datura innoxia. Oxidation of formylphenylacetic acid ethyl ester.
An enzyme system from Datura innoxia roots oxidizing formylphenylacetic acid ethyl ester was purified 38-fold by conventional methods such as (NH4)2SO4 fractionation, negative adsorption on alumina Cy gel and chromatography on DEAE-cellulose. The purified enzyme was shown to catalyse the stoicheiometric oxidation of formylphenylacetic acid ethyl ester to benzoylformic acid ethyl ester and formi...
متن کاملThe cyanogenic substrate for horseradish peroxidase is a conjugated enamine.
We identify the cyanogenic substrate for horseradish peroxidase (HRP) as a conjugated enamine and explore this unusual reaction using alpha-aminocinnamate (RH) as follows. 1) HRP catalyzes the oxidation of RH by O2 (and its peroxidation by H2O2 to form R-R) to produce, simultaneously, CN- and benzaldehyde cyanohydrin. 2) RH is transient and must be generated in situ. The properties of the cyano...

1. 3-Phenylpyruvate caused a dose-related stimulation of insulin release from rat pancreatic islets deprived of exogenous nutrient or incubated in the presence of 5.6 or 8.3 mM-D-glucose. 2. 3-Phenylpyruvate inhibited insulin release evoked by high concentrations of D-glucose (16.7 or 27.8 mM) or 4-methyl-2-oxopentanoate (10.0 mM). This inhibitory effect appeared to be attributable to impairmen...
متن کاملThe expression of MaEXP1, a Melilotus alba expansin gene, is upregulated during the sweetclover-Sinorhizobium meliloti interaction.
Expansins are a highly conserved group of cell wall-localized proteins that appear to mediate changes in cell wall plasticity during cell expansion or differentiation. The accumulation of expansin protein or the mRNA for specific expansin gene family members has been correlated with the growth of various plant organs. Because expansin proteins are closely associated with plant cell wall expansi...
متن کاملRegulation of phenylalanine oxidase synthesis in Proteus mirabilis.
Cells of Proteus mirabilis could oxidize L-phenylalanine to phenylpyruvate only when grown in the presence of a number of amino acids, particularly, L-alanine, L-asparagine, L-glutamate, and L-glutamine. Production of phenylalanine oxidase was slowly lost upon growth in a minimal medium containing ammonium ions as a nitrogen source but was reversed by the addition of casein hydrolysate. Oxidase...
